RSKs were first isolated as ribosomal S6 kinases in Xenopus extracts. With the realisation that rapamycin wipes out ribosomal S6 protein phosphorylation without effect on the RSK (instead inhibiting p70 S6 kinase), S6 is no longer considered a physiological target. Several substrates have been identified including Serum Response Factor and GSK-3. This kinase family is unusual in having two kinase-like domains and is activated by direct phosphorylation by the ERKs .
